应用液氮损伤建立兔动脉粥样硬化易损斑块模型的研究

摘要:

目的 研究应用液氮温控气体损伤动脉血管内皮并结合高脂饮食的方法建立兔动脉粥样硬 化易损斑块模型的方法。方法 24只雄性新西兰兔随机均分成3组,每组8只。实验组在手术后给予液氮 温控气体损伤兔左颈总动脉内膜,并继续给予高脂饲料喂养3个月;假手术组仅手术操作,术后继续高 脂饲料喂养3个月;空白对照组不行任何操作,给与普通饲料喂养3个月。3组动物在处死前均行鲁塞尔 蝰蛇毒+组胺药物触发斑块不稳定,动物在处死后,取左颈总动脉行HE染色后观察血管形态变化,并取 血分析炎症细胞因子白细胞介素-2(IL-2)、白细胞介素-10(IL-10)、肿瘤坏死因子-α(TNF-α) 在三组中的血清学表达。结果 3个月后,与假手术组和空白对照组相比,实验组有明显易损斑块形成, 且炎症因子IL-2、IL-10、TNF-ɑ表达较假手术组和空白对照组均有明显升高,差异均有统计学意义(P <0.05)。结论 液氮温控气体损伤并结合高脂饮食的方法可成功建立动脉粥样硬化易损斑块模型,且与 动脉粥样硬化相关的炎症细胞因子(IL-2、IL-10、TNF-α)表达明显增加。

Abstract:

Objective To study the method of establishing atherosclerotic vulnerable plaques in rabbits by applying liquid nitrogen temperature-controlled gas to damage arterial endothelium combined with high-fat diet. Methods Male New Zealand rabbits (n=24) were randomly divided into 3 groups (each n=8). The test group was given liquid nitrogen temperature-controlled gas to damage rabbits’ intima of left common carotid artery (CCA) after operation and continuously given high-fat forage for 3 months. The sham-operation group was only given operation and continuously given high-fat forage for 3 months. The blank control group was only given ordinary forage for 3 months. Before execution, all rabbits in 3 groups were given Roussel viper venom and histamine drug for inducing vulnerable plaques. After execution, the changes of CCA morphology were observed after HE staining. The expressions of interleukin-2 (IL-2), interleukin-10 (IL-10) and tumor necrosis factor-α (TNF-α) were analyzed in 3 groups. Results After 3 months, there were significant vulnerable plaques formed, and expressions of IL-2, IL-10 and TNF-α increased significantly in test group compared with sham-operation group and blank control group (P<0.05). Conclusion Liquid nitrogen temperature-controlled gas combined with high-fat diet can successfully establish animal model of atherosclerotic vulnerable plaques. The expressions of inflammatory cytokines (IL-2, IL-10 and TNF-α) related to atherosclerotic increase significantly in the model.
