HIF-1α在七氟醚后处理减轻大鼠心肌缺血再灌注致线粒体功能损伤中的作用研究

摘要:

目的 探讨缺氧诱导因子-1α(HIF-1α)在七氟醚后处理减轻大鼠心肌缺血再灌注致线 粒体功能损伤中的作用。方法 取Langendorff离体灌注模型成功的大鼠心脏88例,采用随机数字表法分为 4组(n=22):对照组(C组)、缺血再灌注组(I/R组)、七氟醚后处理组(Spost组)和HIF-1α抑制剂 2ME2组(2ME2组)。Western blot测定HIF-1α表达水平;电镜观察线粒体超微结构;汉莎氧电极法测 定线粒体3态呼吸(State3)、呼吸控制比(RCR);ATP试剂盒检测ATP含量;线粒体膜电位检测试剂 盒检测膜电位变化。结果 与I/R组比较,Spost组HIF-1α表达上调,State3、RCR、ATP、膜电位升高(P <0.05)。结论 HIF-1α表达上调改善线粒体功能是介导七氟醚后处理减轻大鼠心肌缺血再灌注损伤的 重要调控机制。

Abstract:

Objective To investigate the role of hypoxia-inducible factor-1α (HIF-1α) in sevoflurane posttreatment in reducing myocardial ischemia-reperfusion-induced mitochondrial function injury in rats. Methods Eighty-eight rat hearts with Langendorff isolated perfusion model were randomly divided into 4 groups (n=22): control group (C group), ischemia-reperfusion group (I/R group), seven Fluorether post-treatment group (Spost group) and HIF-1α inhibitor 2ME2 group (2ME2 group). Western blot was used to determine the expression level of HIF-1α; mitochondrial ultrastructure was observed by electron microscope; Lufthansa oxygen electrode method was used to measure mitochondrial 3-state respiration (State3) and respiratory control ratio (RCR); Membrane potential changes. Results Compared with the I/R group, HIF-1α expression was up-regulated in the Spost group, and State3, RCR, ATP, and membrane potentials were increased (P<0.05). Conclusion Up-regulation of HIF- 1α expression improves mitochondrial function is an important regulatory mechanism that mediates sevoflurane postconditioning to attenuate myocardial ischemia-reperfusion injury in rats.
